目的:体外观察盐酸川芎嗪、葛根素、丹参酮ⅡA、丹皮酚对血管紧张素Ⅱ(ANG-Ⅱ)诱导的大鼠血管外膜成纤维细胞(VAF)的增殖及胶原蛋白合成的影响。方法:MTT法检测细胞增殖活性;羟脯氨酸比色法测定细胞培养上清液中胶原蛋白含量。结果:4种中药成分能够显著降低ANG-Ⅱ诱导的VAF增殖及细胞培养上清液中羟脯氨酸含量。结论:盐酸川芎嗪、葛根素、丹参酮ⅡA、丹皮酚能够抑制ANG-Ⅱ诱导的VAF增殖和胶原合成。
OBJECTIVE: To observe the effects of tetramethylpyrazine hydrochloride, puerarin, tanshinone IIA, and paeonol on the proliferation and collagen synthesis of rat adventitial fibroblasts (VAF) induced by angiotensin II (ANG-II). Methods: MTT assay was used to detect cell proliferation activity. Hydroxyproline colorimetric method was used to determine collagen content in cell culture supernatant. RESULTS: The four Chinese herbal ingredients could significantly reduce the ANG-II-induced VAF proliferation and the hydroxyproline content in the cell culture supernatant. Conclusion: Tetramethylpyrazine hydrochloride, puerarin, tanshinone IIA and paeonol can inhibit the proliferation and collagen synthesis of VAF induced by ANG-II.
