目的:探讨补肾益髓方药(益髓生血颗粒)对辐射损伤致小鼠肾虚髓损模型的治疗作用。方法:采用60Coγ射线3.5 Gy全身一次性照射,造成小鼠骨髓造血抑制、外周血象有形成分减少,致小鼠肾虚髓损。用益髓生血颗粒高、中、低剂量(含生药23.6,11.8,5.9 g.kg-1,ig)照射前预防给药2 d,造模后再连续给药14 d。在照射前、照射后1,3,5,7,11,13,15,17,21 d检测外周血象。结果:与模型组比较,阳性药粒细胞集落刺激因子(G-CSF)在造模后1~17 d白细胞数显著升高(P<0.05,P<0.01);益髓生血颗粒各剂量组均能升高白细胞,高、中、低剂量组分别在造模后1,5,7,13,17 d,促进白细胞数升高(P<0.05或P<0.01);照射后各组的红细胞、血红蛋白均有一定程度的下降,G-CSF和益髓生血颗粒各剂量组红细胞数在15,17 d高于模型组(P<0.05或P<0.01);模型组血小板计数在照后5 d开始下降,照后7 d达最低值,此后缓慢回升,而用药组动物从照射后5 d起较模型组恢复快,其中高剂量组在5,11,13 d、中剂量在13 d明显高于模型组(P<0.05,P<0.01,P<0.01)。结论:益髓生血颗粒各剂量组均能显著升高辐射损伤致小鼠外周血白细胞数,能促进辐射损伤小鼠外周血红细胞、血红蛋白的恢复,促进照射小鼠外周血血小板数的回升,补肾益髓方药具有补肾填精保护骨髓促进造血作用。
Objective: To investigate the therapeutic effect of Bushen Yisui Recipe (Yisui Shengxue Granule) on the model of kidney deficiency in mice with radiation injury. Methods: Whole-body irradiation with 3.5 Gy of 60Coγ-ray resulted in inhibition of bone marrow hematopoiesis in mice and decreased formation of peripheral blood astrocytoma in mice. The prophylactic administration of Yisheng Shengxue Granule for 2 days before irradiation at high, medium and low doses (including crude drugs 23.6, 11.8, 5.9 g.kg-1, ig) and continuous administration for 14 days after modeling. Before irradiation, 1, 3, 5, 7, 11, 13, 15, 17 and 21 days after irradiation, peripheral blood was detected. Results: Compared with the model group, the number of leukocytes in the positive granulocyte colony-stimulating factor (G-CSF) increased significantly from 1 to 17 days after model establishment (P <0.05, P <0.01) (P <0.05 or P <0.01). The numbers of erythrocyte, erythrocyte, leucocyte, Hemoglobin decreased to a certain extent. The numbers of erythrocytes in G-CSF and Yisheng Shengxue granule were higher than those in the model group at 15 and 17 d (P <0.05 or P <0.01). The platelet count of the model group began at 5 d , And reached the lowest value on the 7th day after irradiation, then rose slowly. However, the animals in the treatment group recovered faster than the model 5 days after irradiation, in which the high dose group was significantly higher than the 5th, 13th and 13th day Model group (P <0.05, P <0.01, P <0.01). Conclusion: Yisui Shengxue Granule can significantly increase the number of peripheral blood leukocytes in radiation-injured mice, promote the recovery of peripheral blood red blood cells and hemoglobin, and promote the recovery of peripheral blood platelets in irradiated mice. Benefit marrow prescriptions have kidney fill to protect the bone marrow to promote hematopoietic function.
