目的观察促肝细胞生长素(PHGF)在成人肝细胞(HH)增殖和细胞周期中的作用。方法体外培养一定时间后,HH分成空白对照组和5个PHGF剂量(10、20、40、80、160)μg/ml干预组,采用台盼蓝染色,计数CCK-8检测细胞增殖,流式细胞术检测细胞周期。结果PHGF对HH增殖有明显促进作用,PHGF20和40μg/ml两组HH增殖明显大于对照组(P<0.05);与对照组相比,PHGF作用48h后,20和40μg/ml两组G0/G1期细胞比例减少,S期G2/M期细胞比例增加(P<0.05)。结论在一定剂量范围内,PHGF促进HH增生,其可能机制是通过促进DNA合成,增加进入S期细胞数目实现的。
Objective To investigate the role of hepatocyte growth-promoting factor (PHGF) in the proliferation and cell cycle of adult hepatocytes (HH). Methods After a certain period of in vitro culture, HH was divided into blank control group and 5 PHGF groups (10, 20, 40, 80, 160) μg / ml. Cell proliferation was detected by trypan blue staining and counting of CCK- Cytometry detects cell cycle. Results Compared with the control group, the proliferation of HH in PHGF20 and 40μg / ml groups was significantly increased (P <0.05). After 48h, the G0 / G1 The proportion of senescent cells decreased, while the proportion of cells in S phase increased (P <0.05). Conclusion In a certain dose range, PHGF promotes HH proliferation. Its possible mechanism is through promoting DNA synthesis and increasing the number of cells entering S phase.
