目的通过观察大鼠肝纤维化形成过程中胰岛素样生长因子-Ⅰ受体(IGF-ⅠR)的动态变化,探讨IGF-ⅠR与肝纤维化的关系。方法制备四氯化碳诱导的大鼠肝纤维化模型,共8周。测定各组门静脉压,对肝组织进行常规苏木精-伊红和马松三色染色,采用免疫组织化学方法测定肝组织IGF-ⅠR的表达,通过图像分析系统进行定量分析,并将IGF-ⅠR与肝纤维化程度、门静脉压进行相关性分析。结果与正常对照组比较,模型组随着造模时间的延长,肝纤维化程度加重,门静脉压逐渐增高,IGF-ⅠR在肝组织中的表达也增加,IGF-ⅠR的表达与肝纤维化程度及门静脉压呈正相关。结论在肝纤维化进程中,IGF-ⅠR表达逐渐增加,提示其可能参与肝纤维化。
Objective To observe the dynamic changes of insulin-like growth factor-Ⅰ receptor (IGF-ⅠR) during the process of hepatic fibrosis in rats and to explore the relationship between IGF-ⅠR and hepatic fibrosis. Methods Carbon tetrachloride-induced rat liver fibrosis model was established for 8 weeks. The portal vein pressure of each group was measured, and the hepatic tissues were routinely stained with hematoxylin-eosin and horse pine trichrome. The expression of IGF-ⅠR in liver tissue was detected by immunohistochemical method. Quantitative analysis was performed by image analysis system. IGF- ⅠR and liver fibrosis, portal pressure correlation analysis. Results Compared with the normal control group, with the extension of modeling time, the degree of liver fibrosis increased, the portal pressure increased gradually and the expression of IGF-ⅠR increased in liver tissue. The expression of IGF-ⅠR and the degree of hepatic fibrosis And portal pressure was positively correlated. Conclusion The expression of IGF-ⅠR in liver fibrosis progressively increased, suggesting that it may be involved in liver fibrosis.
