目的建立蒙古沙冬青中白藜芦醇的高效液相色谱测定方法。方法采用正交试验优化样品处理的提取条件,采用Merk Hibar Lichrospher RP-18e(4.6 mm×250 mm,5μm)色谱柱,流动相24%乙腈,流速1.0 ml·min-1,紫外检测波长为306 nm。结果沙冬青茎中白藜芦醇在0.104 1~6.082 0μg呈良好的线性关系(r=0.999 9),平均回收率为100.31%,RSD为1.36%,叶中未检出白藜芦醇。结论该方法精密、准确、重现性好,可用于沙冬青中白藜芦醇的含量测定。
OBJECTIVE To establish a method for the determination of resveratrol in Astragalus mongolica by high performance liquid chromatography. Methods The orthogonal experiment was used to optimize the extraction conditions of samples. The mobile phase was acetonitrile (24% acetonitrile, flow rate 1.0 ml · min-1) at a wavelength of 306 with a Merk Hibar Lichrospher RP-18e (4.6 mm × 250 mm, nm. Results The results showed that resveratrol had a good linearity (r = 0.999 9) in the range of 0.104 1 ~ 6.082 0 μg with the average recovery of 100.31% and RSD of 1.36%. Resveratrol was not detected in leaves. Conclusion The method is accurate, accurate and reproducible. It can be used for the determination of resveratrol in Astragalus mongolica.
