目的:对温郁金内生真菌活性较高的E8菌株进行菌种鉴定,并对其代谢产物的化学成分进行研究。方法:采用形态学观察结合ITS序列分析法对E8菌株进行了菌种鉴定;应用多种层析手段对其代谢产物的化学成分进行分离和纯化,并依据理化性质及波谱数据鉴定其化学结构。结果:经鉴定菌株E8为青霉属的草酸青霉Penicillium oxalicum Currie&Thom;从菌株E8的代谢产物中分离纯化了4个化合物,分别鉴定为大黄酚(1)、大黄素(2)、黑麦酮酸A(3)和β-谷甾醇(4)。结论:内生真菌草酸青霉P.oxalicum首次从药用植物温郁金中分离;4个化合物均为首次从温郁金内生真菌中分离得到,微生物发酵也是生产这些化合物的一条新途径。
OBJECTIVE: To identify the strains of E8 with high activity of endophytic fungi and to study the chemical constituents of its metabolites. Methods: The strains of E8 were identified by morphological observation and ITS sequence analysis. The chemical constituents of their metabolites were isolated and purified by a variety of chromatographic techniques. Their chemical structures were elucidated on the basis of physico-chemical properties and spectral data. Results: The identified strain E8 was Penicillium oxalicum Currie & Thom of Penicillium. Four compounds were isolated and purified from the metabolites of strain E8 and identified as chrysophanol (1), emodin (2), rutin Acid A (3) and β-sitosterol (4). CONCLUSION: The endophytic fungus P. oxalicum was isolated from medicinal plant Wenyujin for the first time. All four compounds were isolated from endophytic fungi of the genus Typhonium for the first time. Microbial fermentation is also a new way to produce these compounds.
