目的:检测DNA修复基因XRCC1在维吾尔族、汉族乳腺癌组织及癌旁组织中的表达状况,探讨其与乳腺癌发生的关系及种族差异。方法:选取维吾尔族和汉族乳腺癌患者各70例,应用实时荧光定量PCR测定癌组织及癌旁组织中XRCC1 mRNA的表达。其中随机选取维、汉各27例,采用Western blot法检测癌组织及癌旁组织中XRCC1蛋白表达。结果:XRCC1 mRNA在维、汉乳腺癌组织中表达均高于相应癌旁组织(P<0.05),蛋白在维、汉乳腺癌组织中表达均低于相应癌旁组织(P<0.05),在转录水平表现出种族差异(P<0.05)。结论:XRCC1在乳腺癌组织及癌旁组织中存在转录及翻译水平的差异表达,并表现出种族差异性。
OBJECTIVE: To detect the expression of DNA repair gene XRCC1 in Uygur and Han nationality breast cancer tissues and paracancerous tissues, and to explore its relationship with breast cancer and ethnic differences. Methods: Seventy patients with Uighur and Han nationality were enrolled in this study. The expression of XRCC1 mRNA was detected by real-time fluorescence quantitative PCR in cancer tissues and adjacent tissues. Among them, 27 cases were randomly selected from the dimension and the Han, and Western blot was used to detect the expression of XRCC1 protein in cancer tissues and adjacent tissues. Results: The expression of XRCC1 mRNA in both breast cancer tissues was significantly higher than that in adjacent tissues (P <0.05). The protein expression of XRCC1 in both breast cancer tissues was lower than that in corresponding paracancerous tissues (P <0.05) The transcriptional level showed racial differences (P <0.05). Conclusion: The expression of XRCC1 in breast cancer tissues and paracancerous tissues has different transcription and translation levels, and shows racial differences.
