本文介绍用微孔培养板取代培养瓶培养新生大鼠心肌细胞,并用400 TCI_(50) 0.05 ml/孔CoxsackieB_5病毒感染心肌细胞.在感染后1,6、24及36h分别加入水飞蓟宾,观察到用水飞蓟宾后培养液中LDH、GOT均低于病毒对照组,感染后DNA合成率明显高于病毒对照组,以及96 h受感染细胞培养液中病毒滴度(Lg 3.95 TCID_(50))明显低于病毒对照组(Lg 6.95 TCID_(50)).本实验结果提示水飞蓟宾对Coxsackie B_5病毒感染培养的心肌细胞有明显保护作用,尤其是在感染后1~6 h给药,效果更好。
In this paper, the cultured neonatal rat cardiomyocytes were cultured in microporous culture plates instead of culture flasks, and infected with 400 TCI_ (50) 0.05 ml / well Coxsackie B_5 virus.Meanwhile, silybin was added at 1, 6, 24 and 36 h after infection, The results showed that LDH and GOT in the culture fluid after silybin injection were lower than those in the virus control group. The DNA synthesis rate after infection was significantly higher than that in the virus control group, and the virus titer (Lg 3.95 TCID 50 ) Was significantly lower than that of virus control group (Lg 6.95 TCID 50) .The results of this experiment suggest that silibinin has a significant protective effect on cardiomyocytes cultured with Coxsackie B_5 virus infection, especially in 1 ~ 6 h after infection ,Better results.
