NF-κB与IL-8在宫颈癌细胞株中的表达及意义

目的探讨核转录因子NF-κB及炎症相关的CXC趋化因子IL-8在人子宫颈癌细胞株Siha、CaSki中的表达及意义。方法选择两种宫颈癌细胞(未转移的宫颈鳞癌细胞株Siha、宫颈癌肠转移细胞株CaSki细胞)采用细胞免疫化学SV两步法检测两种宫颈癌细胞中NF-κB及IL-8的表达情况;采用酶联免疫吸附试验ELISA法检测两种宫颈癌细胞上清液中IL-8的表达情况,同法检测两种宫颈癌细胞的蛋白提取液中NF-κB的表达情况。结果 NF-κB及IL-8在Siha、CaSki两种宫颈癌细胞中均高表达,且两者在肠转移的CaSki细胞中表达明显高于未转移的宫颈癌细胞Siha细胞,差异有统计学意义(P<0.05)。结论 NF-κB及IL-8在宫颈癌细胞株CaSki中的高表达与宫颈癌的转移及发展密切相关。 Objective To investigate the expression and significance of NF-κB and CXC chemokine IL-8 in human cervical cancer cell lines Siha and CaSki. Methods Two types of cervical cancer cells (Siha cervical cancer cell line without metastasis and CaSki cells cervical metastasis) were used to detect the expression of NF-κB and IL-8 The expression of IL-8 in the two kinds of cervical cancer cell supernatants was detected by enzyme-linked immunosorbent assay (ELISA). The same method was used to detect the expression of NF-κB in the protein extracts from two kinds of cervical cancer cells. Results Both NF-κB and IL-8 were highly expressed in Siha and CaSki cervical cancer cells, and the expression of NF-κB and IL-8 in CaSki cells was significantly higher than that in non-metastasized cervical cancer cells, the difference was statistically significant (P <0.05). Conclusion The high expression of NF-κB and IL-8 in cervical cancer cell line CaSki is closely related to the metastasis and development of cervical cancer.
