目的:研究长梗秦艽(Gentiana waltoniiBurkill)的化学成分,为阐明其药效物质奠定基础。方法:通过大孔树脂、硅胶、sephadex LH-20、反相硅胶(Rp-18)柱层析及制备液相色谱(Prep LC)进行分离,根据化合物理化性质和光谱数据鉴定结构。结果:从该植物中分离鉴定了10个化合物,分别为:gentiolactone(1)、2-methoxy-anofinic acid(2)、4,6-二羟基-2-甲氧基苯乙酮(4,6-dihydroxy-2-methoxyacetophenone,3)、东莨菪素(scopoletin,4)、6′-(2,3-二羟基苯甲酰)当药苷酯(6′-(2,3-dihydroxybenzoyl)sweroside,5)、3′-乙酰基当药苷(3′-acetylsweroside,6)、4-O-2-羟基-6-甲氧基苯乙酮苷(4-O-glycosyloxy-2-hydroxy-6-methoxyacetophenone,7)、獐牙菜苦苷(swertiamarin,8)、当药苷(sweroside,9)和龙胆苦苷(gentiopicroside,10)。结论:化合物3、4和7为首次从龙胆属植物中分离得到,化合物1~10均为首次从该植物中分离得到。
Objective: To study the chemical constituents of Gentiana waltonii Burkill and lay a foundation for clarifying its effective substances. METHODS: Macroporous resin, silica gel, sephadex LH-20, reversed-phase silica gel (Rp-18) column chromatography and preparative LC (Prep LC) were used for separation. The structures were identified according to the physicochemical properties and spectral data of the compounds. Results: Ten compounds were isolated and identified from this plant: gentiolactone (1), 2-methoxy-anofinic acid (2), 4,6-dihydroxy-2-methoxyacetophenone (4,6 -dihydroxy-2-methoxyacetophenone, 3), scopoletin (4), 6’-(2,3-dihydroxybenzoyl)-sweroside, 6’-(2,3-dihydroxybenzoyl)sweroside, 5) 3′-acetylsweroside (6), 4-O-2-glycosyloxy-2-hydroxy-6- Methoxyacetophenone, 7), swertiamarin (8), swerbee (sweroside, 9) and gentiopicroside (10). Conclusion: Compounds 3, 4 and 7 were isolated from Gentianae for the first time. Compounds 1~10 were isolated from this plant for the first time.
