目的建立提高生活饮用水中隐孢子虫和贾第鞭毛虫(两虫)检出率的方法。方法用1.2μm微孔滤膜通过真空抽滤的方式过滤两虫加标样品,后将滤膜置于磷酸缓冲盐溶液(phosphate buffer saline,PBS)中进行超声洗脱,收集洗脱液离心富集卵囊和孢囊;按照GB/T 5750.12-2006《生活饮用水卫生标准检验方法》洗脱液离心富集卵囊和孢囊。将2种试验方法离心收集到的卵囊和孢囊分别进行免疫磁分离和荧光抗体染色,计算2种方法的回收率。结果优化方法对隐孢子虫和贾第鞭毛虫的加标回收率分别为60%和56%,标准方法GB/T 5750.12-2006的加标回收率分别为22%和20%。结论优化方法对于提高生活饮用水中两虫检出率是有效的,且可以达到降低检测成本的目的。
Objective To establish a method to improve the detection rate of Cryptosporidium and Giardia in drinking water. Methods The samples of spiked worms were filtered through a vacuum filtration using a 1.2 μm microporous membrane filter. The membrane was then placed in phosphate buffered saline (PBS) for elution. The eluate was centrifuged Oocysts and cysts were collected; and oocysts and cysts were enriched by centrifugation according to GB / T 5750.12-2006 “Standard Test Method for Drinking Water Quality”. The two methods were centrifuged to collect the oocysts and cysts were immunomagnetic separation and fluorescent antibody staining to calculate the recovery of the two methods. Results The spiked recoveries of Cryptosporidium parvum and Giardia were 60% and 56%, respectively. The standard recoveries of GB / T 5750.12-2006 were 22% and 20%, respectively. Conclusion The optimization method is effective for increasing the detection rate of two insects in drinking water and can reduce the detection cost.
