目的:应用原子力显微镜(AFM)和透射电镜观察低强度电磁场(LIPEMF)对表皮干细胞超微结构的影响。方法:对经LIPEMF作用5d(选取频率50Hz,5mT,30min/次,1次/d)和未经LIPEMF作用的体外培养的人表皮干细胞,用AFM和透射电镜进行细胞超微结构观察。应用AFM,两组分别测量10个表皮干细胞的细胞直径、细胞表面平均粗糙度和均方根粗糙度。结果:从AFM可见,人表皮干细胞表面分布着大量直径几十到几百纳米不等的小颗粒,经LIPEMF作用的表皮干细胞表面的颗粒高度、粗糙度等较未经LIPEMF作用的细胞高;透射电镜发现作用组细胞稍较成熟,核浆比例小,胞质细胞器稍多。结论:低强度电磁场对人表皮干细胞的超微结构有一定影响,AFM有助于观察其表面超微结构变化。
Objective: To observe the effect of low-intensity electromagnetic field (LIPEMF) on the ultrastructure of epidermal stem cells by atomic force microscope (AFM) and transmission electron microscopy (TEM). Methods: Human epidermal stem cells cultured in vitro with LIPEMF for 5 days (frequency 50 Hz, 5 mT, 30 min / time, once / d) and without LIPEMF were observed by AFM and transmission electron microscopy. AFM was used to measure the cell diameter, cell surface average roughness and root mean square roughness of 10 epidermal stem cells in both groups. Results: From the AFM, a large number of small particles ranging in diameter from tens to hundreds of nanometers were distributed on the surface of human epidermal stem cells. The surface height and roughness of epidermal stem cells treated with LIPEMF were higher than those without LIPEMF. Electron microscopy found that the role of the group of cells slightly mature, a small proportion of nuclear plasma, cytoplasmic organelles slightly more. CONCLUSION: Low-intensity electromagnetic fields have some influence on the ultrastructure of human epidermal stem cells. AFM is helpful to observe the ultrastructural changes of human epidermal stem cells.
