目的研究水黄皮根总黄酮(PRF)对大鼠幽门结扎型胃溃疡的作用机制。方法采用大鼠幽门结扎模型观察PRF对胃溃疡的保护作用;幽门结扎收集胃液观察PRF对大鼠胃液分泌和黏液分泌的影响;采用免疫组化染色法观察PRF对幽门结扎大鼠胃黏膜表皮生长因子(EGF)表达的影响;采用放射免疫方法观察PEF对幽门结扎大鼠血清胃泌素的影响。结果与模型对照组的溃疡指数(4.37±0.69)比较,PRF中、高剂量组分别为(2.62±0.69),(2.12±0.92),对大鼠幽门结扎型胃溃疡有明显的保护作用(P<0.01);与模型对照组的积分吸光度(0.67±0.24)比较,PRF低、中、高剂量组分别为(0.93±0.41),(1.36±0.59),(1.54±0.68),差异均有统计学意义(P<0.05,P<0.01);与模型对照组比较,PRF各剂量组可明显降低胃液量(P<0.05,P<0.01);PRF中、高剂量能明显降低大鼠游离酸度、总酸度以及总酸排出量(P<0.01)。结论水黄皮根总黄酮对大鼠幽门结扎型胃溃疡具有明显保护作用,可能与通过抑制胃泌素分泌、促进EGF分泌、促进胃黏液分泌有关。
Objective To study the mechanism of PRF on pyloric gastric ulcer in rats. Methods The rat model of pylorus ligation was used to observe the protective effect of PRF on gastric ulcer. The effect of PRF on secretion of gastric juice and mucus secretion in rats were observed by pylorus ligation. The gastric mucosal epithelial growth was observed by immunohistochemical staining Factor (EGF) expression; using radioimmunoassay method to observe PEF on pylorus ligation rat serum gastrin impact. Results Compared with the ulcer index (4.37 ± 0.69) in model control group, the levels of PRF and high dose group were (2.62 ± 0.69) and (2.12 ± 0.92), respectively, which showed obvious protective effect on pylorus-ligated gastric ulcer <0.01). Compared with the integral absorbance of model control group (0.67 ± 0.24), the levels of PRF low, medium and high dose were (0.93 ± 0.41), (1.36 ± 0.59) and (1.54 ± 0.68) (P <0.05, P <0.01). Compared with the model control group, the levels of gastric juice in PRF groups decreased significantly (P <0.05, P <0.01). The medium and high doses of PRF could significantly reduce the free acidity, Total acidity and total acid output (P <0.01). Conclusion The total flavonoids of Radix et Rhizoma Rhei could obviously prevent gastric ulcer in rats with pylorus ligation, which may be related to the inhibition of gastrin secretion, EGF secretion and gastric mucus secretion.
