目的观察ICCs细胞(interstitial cells of Cajal,ICCs)在糖尿病(diabetes mellitus,DM)大鼠膀胱中的密度分布情况,探讨其在糖尿病膀胱逼尿肌肌条兴奋性改变以及膀胱功能改变中的作用。方法腹腔注射链脲佐菌素(strepto-zotocin,STZ)建立糖尿病SD大鼠模型。12周后,行充盈性膀胱测压,比较DM组大鼠与对照组膀胱残余尿量、最大膀胱容量、最大膀胱压力及膀胱顺应性变化;行离体逼尿肌肌条实验,比较肌条兴奋性与对照大鼠的差异;应用免疫组织荧光染色法检测2组大鼠膀胱ICCs细胞的密度分布情况。结果DM组大鼠膀胱残余尿量[(0.50±0.28)ml]、最大膀胱容量[(1.72±0.73)ml]和膀胱顺应性[(0.04±0.02)ml/cmH2O]较对照组明显增加(P<0.01),而最大膀胱压力明显降低(P<0.05)。DM组逼尿肌肌条收缩频率较对照组明显降低(P<0.05),而且收缩幅度有降低趋势。DM大鼠膀胱ICCs细胞数量较对照组明显减少(P<0.01)。结论DM大鼠膀胱中ICCs细胞数量减少可能是糖尿病性膀胱病逼尿肌肌条兴奋性降低及在体膀胱功能改变的重要原因之一。
Objective To observe the density distribution of interstitial cells of Cajal (ICCs) in the bladder of diabetic mellitus (DM) rats and to explore its role in the excitability of bladder detrusor muscle strips and the changes of bladder function. Methods Diabetic SD rats were established by intraperitoneal injection of strepto-zotocin (STZ). Twelve weeks later, we performed filling cystometry to compare bladder residual urine volume, maximum bladder volume, maximum bladder pressure and bladder compliance in DM group and control group. Excitability and control rats; Immunofluorescence staining was used to detect the density distribution of bladder ICCs in two groups. Results The bladder residual urine volume in DM group was significantly higher than that in control group [(0.50 ± 0.28) ml], [(1.72 ± 0.73) ml] and bladder compliance [(0.04 ± 0.02) ml / cmH2O] <0.01), while the maximum bladder pressure was significantly lower (P <0.05). The contractile frequency of detrusor muscle strips in DM group was significantly lower than that in control group (P <0.05), and the contraction amplitude decreased. The number of bladder ICCs in DM rats was significantly decreased compared with the control group (P <0.01). Conclusion The decreased number of ICCs in the bladder of diabetic rats may be one of the important reasons for the decrease of excitability of diabetic bladder detrusor myoblasts and the change of bladder function in the body.
