目的观察通心络对糖尿病小猪心肌组织中转化生长因子β1(TGF-β1)表达的影响,探讨糖尿病心肌病的发病机制及通心络的保护作用。方法制备糖尿病小型猪模型,随机分为糖尿病心肌病组(DCM)和通心络治疗组(TXL),以正常小猪作为对照组(C)。检测各组小猪的血糖变化情况。观察通心络对糖尿病小猪血流动力学的影响。应用免疫组化技术和Western blot技术观察各组动物心肌组织中TGF-β1的表达。结果通心络对糖尿病小猪的血糖无明显影响,但是可明显增加糖尿病小猪心肌血流。与对照组相比,糖尿病组小猪心肌组织中TGF-β1的表达明显增加,通心络可显著降低TGF-β1的表达。结论通心络可改善糖尿病心肌组织的功能,可能通过降低TGF-β1表达这一途径改善心肌纤维化状态,对糖尿病心肌组织具有保护作用。
Objective To observe the effects of Tongxinluo on the expression of transforming growth factor-β1 (TGF-β1) in diabetic piglets and to explore the pathogenesis of diabetic cardiomyopathy and the protective effect of Tongxinluo. Methods Diabetic mini-pigs were prepared and randomly divided into diabetic cardiomyopathy group (DCM) and Tongxinluo treatment group (TXL), and normal piglets as control group (C). Detection of blood glucose in each group of pigs. To observe the effect of Tongxinluo on hemodynamics in diabetic piglets. The expression of TGF-β1 in myocardium of each group was observed by immunohistochemistry and Western blot. Results Tongxinluo no significant effect on the blood glucose of diabetic piglets, but can significantly increase the blood flow in diabetic piglets. Compared with the control group, the expression of TGF-β1 in diabetic piglets increased significantly, Tongxinluo significantly reduced the expression of TGF-β1. Conclusion Tongxinluo can improve the function of diabetic myocardium, and it may improve myocardial fibrosis state by decreasing the expression of TGF-β1, which may have a protective effect on diabetic myocardial tissue.
