目的探讨炎调方对脓毒症大鼠肺组织核因子-κB(NF-κB)活性及血清高迁移率族蛋白B1(HMGB1)水平变化的影响。方法将雄性SD大鼠随机分为正常组、假手术组、模型组和炎调方组。盲肠结扎穿孔术(CLP)制备脓毒症大鼠模型,于造模后2,8,12,24h处死大鼠,凝胶迁移电泳法(EMSA)检测右肺组织NF-κB活性,双抗体夹心酶联免疫吸附法(ELISA)测定HMGB1水平。结果正常组及假手术组大鼠肺组织NF-κB微弱活化,模型组中,CLP术后2h NF-κB活性开始增强,24h仍有较强的活性;炎调方组与模型组相比,NF-κB活性各时间点均明显减弱。模型组HMGB1水平2h开始升高,炎调方组与模型组相比,HMGB1水平明显降低(P<0.05)。结论炎调方能抑制脓毒症大鼠肺组织NF-κB活性,降低脓毒症大鼠血清HMGB1水平。
Objective To investigate the effects of Yan tang Recipe on the activity of nuclear factor-κB (NF-κB) and the level of serum high mobility group box 1 (HMGB1) in lung tissue of septic rats. Methods Male Sprague-Dawley rats were randomly divided into normal group, sham operation group, model group and Yan Tiao Fang group. The model of sepsis was prepared by cecal ligation and puncture (CLP). Rats were sacrificed at 2, 8, 12 and 24 hours after model establishment. The activity of NF-κB in right lung was detected by EMSA. The level of HMGB1 was determined by enzyme-linked immunosorbent assay (ELISA). Results NF-κB in lung tissue of normal rats and sham-operated rats was weakly activated. In the model group, the activity of NF-κB began to increase at 2h after CLP, and still remained strong at 24h. Compared with model group, NF-κB activity significantly decreased at all time points. The level of HMGB1 in model group began to increase at 2h, and the level of HMGB1 in Yan-Fang prescription group was significantly lower than that in model group (P <0.05). Conclusion Yan-tang prescription can inhibit NF-κB activity in lung tissue of septic rats and decrease the level of serum HMGB1 in septic rats.
