目的:观察青蒿琥酯对腺样囊性癌耐顺铂细胞(NACC/DDP)裸鼠腮腺移植瘤的抑瘤效果,探讨其作为抗癌药物的临床应用前景。方法:选用5周龄雄性BALB/C鼠建立NACC/DDP裸鼠腮腺移植瘤模型,随机将裸鼠分成生理盐水对照组、DDP 2 mg/kg组、ART 200 mg/kg组、联合用药组(ART 100 mg/kg加DDP1mg/kg)共4组,隔日腹腔注射给药并测量各组裸鼠体重、移植瘤长短径。给药15次后取瘤,称瘤重,计算各用药组的抑瘤率,苏木精-伊红染色观察各组肿瘤组织病理改变并应用免疫组织化学方法检测Survivin在移植瘤组织中的表达情况。结果:给药4周后DDP 2.0mg/kg组的抑瘤率最高为59.86%(P<0.01),但该组裸鼠明显消瘦,状态不佳;ART 200mg/kg组的抑瘤率为43.54%(P<0.05),该组裸鼠精神状态良好,体重稳定增加;联合用药组的抑瘤率为41.16%(P<0.05),体重稍有减轻。苏木精-伊红染色结果显示:ART 200mg/kg、DDP 2mg/kg组和联合用药组肿瘤细胞核深染、固缩,核染色质浓缩、碎裂成片状,呈细胞凋亡状态,细胞间可见少量红染、无结构的坏死物。结论:青蒿琥酯能够抑制腺样囊性癌耐药细胞(NACC/DDP)在裸鼠体内的增殖并促进其凋亡,且青蒿琥酯对裸鼠的毒副作用较顺铂小。
Objective: To observe the anti-tumor effect of artesunate on parotid gland transplantation tumor in nude mice with adenoid cystic carcinoma and cisplatin-resistant cells (NACC / DDP), and to explore its clinical application prospect as anticancer drug. Methods: Nude mice were randomly divided into normal saline control group, DDP 2 mg / kg group, ART 200 mg / kg group and combination therapy group (n = ART 100 mg / kg plus DDP 1 mg / kg) for 4 groups. The mice were injected intraperitoneally every other day and the body weight of nude mice and the length and breadth of the tumor were measured. The tumors were weighed and the tumor weight was calculated 15 times after administration. The tumor inhibition rate of each drug group was calculated. The hematoxylin-eosin staining was used to observe the histopathological changes in each group and the expression of Survivin in the tumor tissue was detected by immunohistochemistry Happening. Results: The inhibition rate of DDP 2.0mg / kg group was 59.86% (P <0.01) after 4 weeks of administration, but the nude mice in this group were obviously wasting and in poor condition. The inhibition rate of ART in 200mg / kg group was 43.54 % (P <0.05). The nude mice in this group had a good mental state and a steady increase in body weight. The inhibition rate of the combination group was 41.16% (P <0.05), and the body weight was slightly relieved. The hematoxylin-eosin staining results showed that the tumor nuclei of ART 200mg / kg, DDP 2mg / kg group and combination group were deep-stained, condensed and chromatin condensed, fragmented into flake, showing apoptotic state, Visible between a small amount of red dye, no structural necrosis. CONCLUSION: Artesunate can inhibit the proliferation of adenoid cystic carcinoma cell line (NACC / DDP) in nude mice and promote its apoptosis. Artesunate has less side effects than cisplatin on nude mice.
