本文运用半固体琼脂培养法及 MTT法研究了红藻藻蓝蛋白 (R- PC)对 HL- 60细胞生长的影响 ,结果发现 :R- PC能够显著抑制细胞的生长 ,且存在浓度效应和时间效应关系。为进一步探索 R- PC抑制 HL- 60细胞生长的可能机制 ,将不同浓度的 R- PC与 HL- 60细胞相互作用 6d后 ,分别用流式细胞仪检测 HL- 60细胞所处细胞周期中的时相和免疫印渍法测定 HL- 60细胞中 Bcl- 2基因表达产物的变化 ,实验结果显示 :R- PC确能使更多的 HL- 60细胞滞留于细胞周期的 G0 - G1 期 ,但与 R- PC浓度无正向关系 ,且 HL- 60细胞中 Bcl- 2基因表达产物的量也没有明显的变化。结果表明 :R- PC可能通过不同于 G0 - G1 期滞留及 Bcl- 2基因表达的机制抑制 HL- 60细胞的生长。
In this paper, semi-solid agar culture method and MTT method were used to study the effect of phycocyanin (R-PC) on the growth of HL-60 cells. The results showed that: R-PC can significantly inhibit cell growth, and there is concentration effect and time. Effect relationship. In order to further explore the possible mechanism of R-PC inhibiting the growth of HL-60 cells, 6-day interaction between different concentrations of R-PC and HL-60 cells was used to detect the cell cycle of HL-60 cells by flow cytometry. The changes of Bcl-2 gene expression products in HL-60 cells were determined by phase and immunoblot assay. The results showed that R-PC can actually retain more HL-60 cells in the G0-G1 phase of cell cycle, but There was no positive relationship with R-PC concentration and there was no significant change in the amount of Bcl-2 gene expression product in HL-60 cells. The results showed that R-PC may inhibit the growth of HL-60 cells through a mechanism different from that of G0-G1 phase retention and Bcl-2 gene expression.
