目的:考察葛根总黄酮-黄连总生物碱3∶8配伍防治2型糖尿病的作用机制。方法:建立链脲佐菌素(STZ)复合高脂饲料致2型糖尿病大鼠模型,给药后检测血清还原型谷胱甘肽(GSH)、超氧化物歧化酶(SOD)、丙二醛(MDA)、一氧化氮合成酶(NOS)、一氧化氮(NO)水平;免疫组化检测大鼠胰腺组织BAX、BCL-2、Caspase-3蛋白表达。结果:葛根黄连有效组分能显著提高2型糖尿病大鼠血清GSH、SOD活性,降低血清MDA、NOS、NO水平,并能显著提高胰腺组织BCL-2蛋白表达,降低胰腺组织BAX与Caspase-3蛋白表达。结论:“葛根总黄酮-黄连总生物碱”组分配伍对2型糖尿作用机制可能为减轻氧化应激损伤,抑制胰岛β细胞凋亡有关。
OBJECTIVE: To investigate the mechanism of total flavonoids of Radix Puerariae - Coptis total alkaloids with compatibility of 3: 8 in prevention and treatment of type 2 diabetes mellitus. Methods: A rat model of type 2 diabetes induced by streptozotocin (STZ) compound high fat diet was established. Serum reduced glutathione (GSH), superoxide dismutase (SOD), malondialdehyde (MDA), nitric oxide synthase (NOS) and nitric oxide (NO) were detected by immunohistochemical method. The expressions of BAX, BCL-2 and Caspase-3 in pancreatic tissue were detected by immunohistochemistry. Results: The Pueraria rhizome active ingredients can significantly increase serum GSH and SOD activity, reduce serum levels of MDA, NOS and NO in type 2 diabetic rats, and significantly increase the expression of BCL-2 in pancreatic tissues, and decrease the expressions of BAX and Caspase-3 in pancreatic tissues Protein. Conclusion: The combination of “Pueraria Total Flavone-Coptidis Alkaloid” component on type 2 diabetes mellitus may be related to the reduction of oxidative stress injury and the inhibition of pancreatic β-cell apoptosis.
