目的制备丹参酮ⅡA纳米囊,并评价其性能。方法采用乳化-溶剂挥发法制备丹参酮ⅡA纳米囊,以包封率为指标,单因素试验结合正交试验优化制备工艺与处方,并评价纳米囊的粒径形态。结果以乙酸乙酯-水(1∶2)、0.5 mL司盘-80、丹参酮ⅡA-壳聚糖(1∶5)、乙醇0.4 mL为最佳处方,在温度为50℃,搅拌速度为300 r/min条件下乳化,然后超声30 s,即得纳米囊。所得纳米囊外观圆整,平均粒径(172±10)nm,包封率高达73.2%。结论乳化-溶剂挥发法适用于制备丹参酮ⅡA纳米囊,且工艺稳定。
Objective To prepare tanshinone Ⅱ A nanocapsules and evaluate their properties. Methods The tanshinone ⅡA nanocapsules were prepared by emulsion - solvent evaporation method. The entrapment efficiency was used as an index. The single factor experiment and orthogonal experiment were used to optimize the preparation process and prescription. The nanoshells’ size and shape were also evaluated. Results The best prescription was ethyl acetate-water (1: 2), 0.5 mL Span-80, tanshinoneⅡA-chitosan (1: 5) and ethanol 0.4 mL. The temperature was 50 ℃ and the stirring speed was 300 r / min under the conditions of emulsification, and then sonicated 30 s, that is, nanocapsules. The obtained nanocapsules had a round appearance and an average particle size of (172 ± 10) nm with entrapment efficiency as high as 73.2%. Conclusion Emulsification - solvent evaporation method is suitable for the preparation of tanshinone Ⅱ A nanocapsules, and the process is stable.
