目的:探讨蝙蝠葛碱(DAU)对阿尔茨海默病(AD)小鼠学习记忆及海马白介素-12(IL-12)的影响。方法:小鼠随机分成对照组、模型组、吡拉西坦组(0.62 g/kg/d)、DAU高剂量组(1.17 g/kg/d)、DAU低剂量组(0.59 g/kg/d)。采用Aβ_(1-42)双侧脑室注射小鼠诱导AD模型。给予DAU灌胃,治疗18d后取材。应用Morris水迷宫检测小鼠行为学指标及双抗体夹心法检测海马IL-12的水平。结果:与模型组比较,DAU能改善AD模型小鼠的学习和记忆(P<0.05),降低海马IL-12水平(P<0.05)。结论:DAU能改善AD模型小鼠学习记忆功能,DAU通过抑制IL-12在AD治疗中发挥重要作用。
Objective: To investigate the effects of dauricum on learning and memory and the level of interleukin-12 (IL-12) in hippocampus of Alzheimer’s disease (AD) mice. Methods: The mice were randomly divided into control group, model group, piracetam group (0.62 g / kg / d), DAU high dose group (1.17 g / kg / d), DAU low dose group ). AD model was induced by intracerebroventricular injection of Aβ 1-42 in mice. Giving DAU gavage, after 18 days of treatment drawn. The level of IL-12 in the hippocampus was detected by the Morris water maze test and the double antibody sandwich method. Results: Compared with model group, DAU could improve the learning and memory of AD model mice (P <0.05) and decrease the level of IL-12 in hippocampus (P <0.05). Conclusion: DAU can improve learning and memory in AD mice. DAU plays an important role in the treatment of AD by inhibiting IL-12.
