采用液滴法和注射法,将带有苏云金芽孢杆菌毒素蛋白基因的质粒DNA导入自花授粉后的大豆品种鲁豆4号和861387—2。提取D1代大豆叶片的DNA,与苏云金芽孢杆菌毒素蛋白基因进行了斑点杂交,从D1代440个植株中,选出7个杂交阳性后代,表明苏云金芽孢杆菌毒素蛋白基因已整合于大豆基因组中。本文还对外源DNA导入后代筛选中存在的问题进行了讨论。
Plasmid DNA with the Bacillus thuringiensis toxin protein gene was introduced into self-pollinated soybean varieties Luodou No. 4 and No. 861387-2 using droplet method and injection method. The DNA of D1 soybean was extracted and hybridized with the Bacillus thuringiensis toxin protein gene. Seven hybrid positive progeny from 440 plants of D1 generation were selected, indicating that the Bacillus thuringiensis toxin protein gene has been integrated into the soybean genome. This article also discusses the problems existing in the screening of foreign DNA introduction progenies.
