用DNA电泳和荧光染色法流式细胞仪分析,发现在依赖美洲商陆凝集素(PWM)的单核细胞介导的细胞毒活性中,白血病靶细胞U937和Raji细胞均呈现以DNA断裂为特征的细胞程序性死亡。通过细胞毒试验和荧光染色分析表明:抗肿瘤坏死因子α的单克隆抗体和乙酰基葡萄糖胺单糖可分别阻断单核细胞所诱导的U937细胞或Raji细胞死亡和DNA断裂。试验中还发现PWM同时可与单核细胞和这两种肿瘤细胞相结合。提示凝集素在促进效靶细胞相互接近,进而导致肿瘤细胞发生程序性死亡方面具有重要作用。
Using DNA electrophoresis and fluorescence staining, it was found that leukemia target cells U937 and Raji cells were characterized by DNA cleavage in monocyte-mediated cytotoxic activity that relied on the pokeweed (Pooledae, Of programmed cell death. Cytotoxicity assay and fluorescent staining analysis showed that anti-tumor necrosis factor α monoclonal antibody and acetylglucosamine monosaccharide can block monocyte-induced U937 or Raji cell death and DNA breakage, respectively. The experiment also found that PWM can be combined with both monocytes and these two tumor cells. It is suggested that lectins play an important role in promoting target cells close to each other and leading to programmed cell death in tumor cells.
