目的 :探讨呼吸道合胞病毒 (RSV)感染对体外培养人肺腺癌细胞系PAa的影响。方法 :应用RSV体外感染PAa细胞 ,观察PAa细胞的变化 ;聚合酶链反应 (PCR)结合单链构象多态性分析 (SSCP)检测PAa细胞 p5 3基因突变情况。 结果 :RSV感染PAa细胞 2 4h后 ,细胞出现融合病变 ,形成巨融合细胞 ;病变细胞p5 3第 5、6、7、8、9外显子突变热点区域未见单链多态性变化。结论 :RSV对PAa细胞形态影响明显 ,对PAa细胞基因水平的改变有待深入研究。
Objective: To investigate the effect of respiratory syncytial virus (RSV) infection on human lung adenocarcinoma cell line PAa in vitro. Methods: The PAa cells were infected with RSV in vitro and the changes of PAa cells were observed. The mutation of p53 gene in PAa cells was detected by polymerase chain reaction (PCR) and single-strand conformation polymorphism (SSCP). Results: After 24 h of RSV infection, the fusion cells were found in fusion cells and formed giant fusion cells. No single-stranded polymorphism was found in the hot spots of the fifth, seventh, seventh exons of p5 3. Conclusion: The effect of RSV on PAa cell morphology is obvious. The change of PAa cell gene level needs to be further studied.
