目的探讨Wnt信号传导通路在子宫内膜癌侵袭、转移中作用机制,并结合uPA蛋白在子宫内膜癌中的表达并分析二者之间的相关性。方法采用RT-PCR技术,对10例手术切除的新鲜子宫内膜癌和手术切除的新鲜子宫内膜组织进行Wnt-1和uPA mRNA的检测;采用免疫组化SP法对89例子宫内膜癌和30例正常子宫内膜组织中Wnt-1和uPA蛋白的表达进行检测。结果 Wnt-1的高表达与肿瘤的手术分期和淋巴结转移相关(P<0.05);uPA与手术分期、肌层浸润和淋巴结转移相关,具有统计学意义(P<0.05),且Wnt-1在子宫内膜癌组织中阳性表达与uPA的高表达之间呈正相关关系(r=0.434,P<0.05)。结论 Wnt-1和uPA在子宫内膜癌中的表达存在着相关性,推测Wnt-1异常表达通过下游uPA的过度表达促进子宫内膜癌细胞黏附性下降,间质降解和转移。
Objective To explore the mechanism of Wnt signaling pathway in invasion and metastasis of endometrial carcinoma and to analyze the correlation between the expression of uPA protein and endometrial carcinoma. Methods RT-PCR was used to detect the expression of Wnt-1 and uPA mRNA in 10 cases of fresh endometrial carcinoma and fresh endometrial tissue. 89 cases of endometrial carcinoma And 30 cases of normal endometrial tissue Wnt-1 and uPA protein expression were detected. Results The high expression of Wnt-1 was correlated with tumor stage and lymph node metastasis (P <0.05). The level of uPA was correlated with stage, myometrial invasion and lymph node metastasis (P <0.05) There was a positive correlation between the positive expression of uPA and the high expression of uPA in endometrial carcinoma tissues (r = 0.434, P <0.05). Conclusions There is a correlation between the expression of Wnt-1 and uPA in endometrial carcinoma. It is speculated that the abnormal expression of Wnt-1 may promote the adhesion of endometrial carcinoma cells and the degradation and metastasis of endometrial carcinoma by overexpression of downstream uPA.
