用国产抗肿瘤药物三尖杉酯碱处理HL60 和L12 10 细胞 ,探讨药物对两种不同的白血病细胞以及在不同条件下培养的同种白血病细胞核DNA结构的影响。DNA凝胶电泳结果表明 :(1) 0 2 μg/mlHT作用 2h的HL60 细胞和HT作用 2~ 2 4h的L12 10 细胞有明显的DNAladder。 (2 )HT处理 6~ 2 4h的HL60 细胞、在DBA/ 2纯种小鼠体内培养的被HT处理不同时间的L12 10 细胞及所有对照细胞无DNAladder。 (3)HT处理 2 4h的HL60 细胞和HT处理 12h的白血病小鼠体内的L12 10 细胞核DNA断裂成碎片 ,在琼脂糖凝胶上呈弥散状分布。
HL60 and L1210 cells were treated with domestic anti-tumor drug harringtonine to investigate the effect of drugs on DNA structure of two different leukemia cells and allogeneic leukemia cells cultured under different conditions. The results of DNA gel electrophoresis showed that: (1) HL60 cells treated with 0 2 μg / ml HT for 2 hours and HT12-1 cells treated with HT for 24 hours had obvious DNAladder. (2) HT60 cells treated with HT for 6 ~ 24 hours had no DNAladder in L1210 cells and all the control cells treated with HT at different time in DBA / 2 mice. (3) The nuclear DNA of L12 10 cells in HT60 cells treated with HT for 24 hours and in the leukemia mice treated with HT for 12 hours were fragmented and distributed in a diffuse manner on the agarose gel.
