背景:补肾是治疗神经系统老化的主要方法,而神经系统老化主要表现为神经细胞的密度减少、细胞老化和胞质中脂褐素颗粒沉积。目的:探讨滋肾健脑法对老龄小鼠脑海马CA1,CA3区神经细胞密度及超微结构的影响,并与阳性药物维生素E进行对照。设计:完全随机对照实验。单位:湖南省中医药研究院。材料:实验于2000-09/2003-11在湖南省中医药研究院中药研究所药理实验室完成。选择昆明种小鼠30只,益肾健脑1号药液(以下简称滋肾方),由湖南省中医药研究院制剂研究室提供(由制首乌15g熏桑椹15g,枸杞子15g熏五味子6g熏丹参30g熏葛根30g熏红花10g熏石菖蒲10g熏郁金10g熏远志10g熏山楂10g熏全蝎等药物组成,先将全蝎粉碎成细粉,余药加水煎煮2次,合并煎煮液,滤过,加入全蝎细粉即得),生药量为1.0g/mL。方法:30只小鼠随机分为3组,每组10只。对照组给蒸馏水,滋肾方组给滋肾方20.0g/kg,维生素E组给维生素E40.0mg/kg。均灌胃给药,体积为20mL/kg,1次/d,连续给药4周。末次给药后1h,在麻醉状态下迅速处死小鼠。剪开颅骨,取出海马组织,在显微镜下计量CA1和CA3区的神经细胞数,计算出神经细胞密度值。主要观察指标:①各组小鼠脑海马神经细胞密度。②各组小鼠脑海马神经细胞超微结构。结果:30只小鼠均进入结果分析。①脑海马CA1和CA3区神经细胞密度:滋肾方组和维生素E组均高于对照组眼CA1区:(3707±495)熏穴3812±281雪熏穴257±372雪mm-2;CA3区:(2746±262)熏穴2397±366雪熏穴1992±307雪mm-2熏t=2.68~8.30,P<0.05~0.01演。②滋肾方组和维生素E组能减少线粒体老化及胞质中脂褐素颗粒沉积和溶酶体等老化改变。结论:滋肾健脑颗粒对老龄小鼠脑海马区神经细胞密度减少和细胞老化有一定延缓作用。
Background: Bushen is the main method to treat the aging of the nervous system, while the aging of the nervous system is mainly manifested by the decrease of the density of the nerve cells, the cell aging and the deposition of lipofuscin granules in the cytoplasm. Objective: To investigate the effects of nourishing kidney and nourishing the brain on the density and ultrastructure of neurons in hippocampal CA1 and CA3 of hippocampus in aged mice and to compare with the positive drug vitamin E. Design: Complete randomized controlled experiment. Unit: Hunan Provincial Institute of Traditional Chinese Medicine. MATERIALS: Experiments were performed at the Pharmacological Laboratory, Institute of Chinese Materia Medica, Hunan Academy of Traditional Chinese Medicine from September 2000 to November 2003. Select 30 Kunming mice, Yishenjiannao 1 (hereinafter referred to as Zishen Fang), provided by the Hunan Provincial Institute of Traditional Chinese Medicine Preparation Research Center (from the Shouwu 15g smoked mulberry 15g, medlar 15g smoked Schisandra 6g smoked Salvia 30g smoked Pueraria 30g smoked safflower 10g smoked Shichang 10g smoked turban 10g smoked Polygala 10g smoked hawthorn 10g smoked scorpion and other drugs, the first Scorpio crushed into fine powder, the remaining medicine boiling water twice, the merger Decoction, filtration, adding Scorpio powder that), crude drug amount of 1.0g / mL. Methods: Thirty mice were randomly divided into 3 groups with 10 mice in each group. The control group to distilled water, Zishen Fang to Zishen 20.0g / kg, vitamin E group to vitamin E40.0mg / kg. All intragastric administration, the volume of 20mL / kg, 1 / d, continuous administration for 4 weeks. One hour after the last administration, mice were sacrificed immediately under anesthesia. The skull was cut off, the hippocampus was removed, and the number of nerve cells in the CA1 and CA3 areas was counted under a microscope to calculate the nerve cell density value. MAIN OUTCOME MEASURES: ①Nerve cell density in hippocampus of mice in each group. ② Ultrastructure of hippocampal neurons in mice in each group. Results: All 30 mice entered the result analysis. ①Nerve cell density in hippocampal CA1 and CA3 areas: Zishenfang group and vitamin E group were higher than those in the control eyes CA1 area: (3707 ± 495) smoked hole 3812 ± 281 snow smoked hole 257 ± 372 snow mm-2; CA3 District: (2746 ± 262) smoked 2397 ± 366 smoked Xue 1992 ± 307 snow mm-2 smoked t = 2.68 ~ 8.30, P <0.05 ~ 0.01 acting. ② Zishen Fang group and vitamin E group can reduce mitochondria aging and cytoplasmic lipofuscin particles deposition and lysosomal aging changes. Conclusion: Zishenjiannao Granule can delay the degeneration of neurons and cell aging in the hippocampus of aged mice.
