目的:观察益气活血方对放射性肺损伤大鼠血清及肺组织内皮素-1(ET-1)、γ-干扰素(γ-IFN)表达的影响。方法:将66只Wistar大鼠随机分为照射加益气活血方组(益气活血方组)30只、单纯照射组(模型组)30只、正常对照组(对照组)6只。于受照当天开始,益气活血方组每天予益气活血中药10 g.kg-1,ig,模型组及对照组予蒸馏水10 g.kg-1,ig,直至动物被处死取材。益气活血方组与模型组均于首次照射后第4,6,8,12,26周末各取6只,对照组6只于第8周末处死,均取血清及右肺组织,检测ET-1,γ-IFN的表达。结果:益气活血方组及模型组ET-1的肺组织表达及血清值均于照射后第4周升高,8周达到高峰,26周下降,益气活血方组各时相ET-1的表达肺组织及血清值均低于模型组(P<0.05)。而γ-IFN值于第4周开始降低,12周至低谷,26周回升。益气活血方组γ-IFN的表达肺组织及血清值均高于模型组(P<0.05)。结论:益气活血方能调控ET-1,γ-IFN的表达及分泌,对放射性肺损伤具有较好的防治作用。
Objective: To observe the effect of Yiqi Huoxue Fang on the expression of ET-1 and IFN-γ in serum and lung tissue of rats with radiation-induced lung injury. Methods: Thirty - six Wistar rats were randomly divided into three groups: irradiation plus Yiqi Huoxue prescription group (30 cases), irradiation group (model group) and control group (control group). On the day of irradiation, Yiqi Huoxue prescription group was treated with Yiqi Huoxue Chinese medicine 10 g.kg-1 daily, ig, model group and control group with distilled water 10 g.kg-1, ig, until the animals were sacrificed. Six of them were taken on the 4th, 6th, 8th, 12th and 26th week after the first irradiation in the Yiqi Huoxue recipe group and the model group, respectively. Six rabbits in the control group were sacrificed at the end of the eighth week. Serum and right lung tissue were taken for ET- 1, γ-IFN expression. Results: The expression of ET-1 and the serum level of ET-1 in Yiqi Huoxue prescription group and model group were both increased at the 4th week after irradiation, peaked at 8 weeks and decreased at 26 weeks. ET-1 The expression of lung tissue and serum were lower than the model group (P <0.05). The γ-IFN value began to decrease in the fourth week, 12 weeks to the trough, 26 weeks recovery. The expression of γ-IFN in Yiqi Huoxue prescription group was higher than that in model group (P <0.05). Conclusion: Yiqi Huoxue Fang can regulate the expression and secretion of ET-1 and γ-IFN, and has good preventive and therapeutic effects on radiation-induced lung injury.
