目的:研究子宫内膜癌组织中PTEN、诱导型一氧化氮合酶(iNOS)及环氧合酶-2(COX-2)蛋白的表达,以及PTEN、iNOS及COX-2表达与临床病理的关系,探讨三者之间的相关性。方法:采用免疫组化EnVisionTM法检测55例子宫内膜腺癌,24例子宫内膜不典型增生和20例正常子宫内膜石蜡切片中PTEN、iNOS及COX-2的表达。结果:(1)子宫内膜癌组织与子宫内膜不典型增生、正常子宫内膜组织比较,PTEN阳性表达率明显降低,差异有显著性(P<0.05);iNOS、COX-2表达率明显增加,差异有显著性(P<0.05)。(2)PTEN表达与病理分级、肌层浸润程度、淋巴结转移有关(P<0.05);随肿瘤子宫肌层浸润深度的增加,iNOS的表达显示逐渐增强(P<0.05)。(3)在子宫内膜癌组织中,iNOS与PTEN表达呈负相关(r=-0.212,P<0.05),与COX-2表达呈正相关(r=0.513,P<0.05)。结论:PTEN的失表达与iNOS、COX-2的过表达在子宫内膜癌的发生发展中起着一定的作用,PTEN与iNOS、COX-2的异常表达密切相关,三者的联合检测有助于子宫内膜癌的早期诊断。
Objective: To investigate the expression of PTEN, iNOS and COX-2 in endometrial carcinoma and the correlation between the expression of PTEN, iNOS and COX-2 and the clinicopathological parameters Relationship, explore the correlation between the three. Methods: The expressions of PTEN, iNOS and COX-2 in 55 cases of endometrial adenocarcinoma, 24 cases of endometrial dysplasia and 20 cases of normal endometrial paraffin sections were detected by EnVisionTM immunohistochemistry. Results: (1) The expression of PTEN in endometrial carcinoma was significantly lower than that in endometrial atypical hyperplasia and normal endometrium (P <0.05). (2) PTEN expression was correlated with pathological grade, myometrial invasion and lymph node metastasis (P <0.05). The expression of iNOS increased with the depth of myometrial invasion (P <0.05). (3) There was a negative correlation between iNOS and PTEN expression in endometrial carcinoma (r = -0.212, P <0.05), which was positively correlated with COX-2 expression (r = 0.513, P <0.05). CONCLUSION: The loss of PTEN expression and the overexpression of iNOS and COX-2 play an important role in the carcinogenesis and progression of endometrial carcinoma.
