目的探讨脐血红细胞调控白细胞CXCR4表达能力的变化。方法采用灭活癌细胞(S180,5×106/ml)0·2ml(或生理盐水)加入全血细胞(或白细胞)0·2ml和血浆0·3ml中,37℃温育1h,用流式细胞仪免疫荧光法测定红细胞CD35和白细胞(淋巴细胞、粒细胞)CXCR4分子表达量。分组:①癌细胞悬液0·2ml加全血细胞悬液0·2ml和血浆0·3ml;②生理盐水0·2ml加全血细胞悬液0·2ml和血浆0·3ml;③癌细胞悬液0·2ml加白细胞悬液0·2ml和血浆0·3ml;④生理盐水0·2ml加白细胞悬液0·2ml和血浆0·3ml。结果脐血红细胞CD35表达量在S180激活后下降不如正常人红细胞明显,而白细胞CXCR4表达量在S180激活后上升不明显;正常人全血红细胞CD35表达量在S180激活后明显下降(40·21%±11·52%vs28·65%±8·08%,P<0·01),而粒细胞CXCR4表达量明显上升(40·62%±17·89%vs64·18%±11·69%,P<0·01)。结论脐血红细胞免疫调控能力和白细胞免疫功能都明显低下。
Objective To investigate the changes of cord blood erythrocyte regulation of leukocyte CXCR4 expression. Methods To inoculate 0.2 ml of whole blood cells (or leukocytes) and 0.3 ml of plasma with inactivated cancer cells (S180,5 × 106 / ml), 0.2 ml (or normal saline) and incubate at 37 ℃ for 1 hour, The expression of CXCR4 on erythrocytes CD35 and leukocytes (lymphocytes and granulocytes) was measured by immunofluorescence. Grouping: ① cancer cell suspension 0.2 ml plus whole blood cell suspension 0.2 ml and plasma 0.3 ml; ② saline 0.2 ml plus whole blood cell suspension 0.2 ml and plasma 0.3 ml; ③ cancer cell suspension 0 · 2ml plus white blood cell suspension 0 · 2ml and plasma 0 · 3ml; ④ saline 0 · 2ml leukocyte suspension 0 · 2ml and plasma 0.3ml. Results The expression of CD35 in umbilical cord blood erythrocytes decreased less after S180 activation than that of normal human erythrocytes, while the expression of CXCR4 in leucocytes did not increase significantly after S180 activation. The expression of CD35 in normal human erythrocytes decreased significantly after activation of S180 (40.21% ± 11 · 52% vs 28 · 65% ± 8.08%, P <0.01), while the expression of CXCR4 in granulocytes increased significantly (40.62 ± 17.89% vs64.18 ± 11.69% P <0.01). Conclusion The umbilical cord erythrocyte immunoregulatory capacity and leukocyte immune function are significantly lower.
