目的:研究北柴胡正丁醇萃取部位对小鼠急性肝损伤的保护作用并对其化学成分特征进行初步研究。方法:采用腹腔注射四氯化碳诱导小鼠急性肝损伤,以柴胡正丁醇萃取部位高、中、低剂量(20,10,4 g.kg-1)同时预防给药7 d,检测空白组、模型组、对照组、不同剂量柴胡正丁醇萃取部位组小鼠血清丙氨酸转氨酶(ALT),天冬氨酸转氨酶(AST)活性,同时测定肝组织中丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性;建立并分析北柴胡正丁醇萃取部位HPLC色谱图。结果:北柴胡正丁醇萃取部位是以柴胡皂苷a和柴胡皂苷d为主要成分的柴胡总皂苷,可有效抑制造模小鼠血清中ALT,AST的升高(P<0.05),同时可提高造模小鼠肝组织中SOD的活力,降低肝组织中MDA的含量(P<0.05)。结论:北柴胡正丁醇萃取部位对于小鼠急性肝损伤具有保护作用,其化学成分以柴胡皂苷类为主。
OBJECTIVE: To study the protective effect of Bupleurum n-Butyl alcohol extract on acute liver injury in mice and its chemical composition. Methods: Acute hepatic injury induced by intraperitoneal injection of carbon tetrachloride in mice was induced by intraperitoneal injection of 20, 10 and 4 g.kg-1 of Bupleurum n-butanol at 7, , The model group and the control group. The levels of serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST), and the content of malondialdehyde (MDA) Oxidative dismutase (SOD) activity; HPLC chromatogram of n-butanol extraction part of Bupleurum chinense was established and analyzed. Results: The extract fraction of n-butanol from Bupleurum chinense Bupleurum total saponin with saikosaponin a and saikosaponin d as the main components could effectively inhibit the increase of serum ALT and AST (P <0.05) The activity of SOD in the liver of model mice can be increased and the content of MDA in liver tissue can be decreased (P <0.05). Conclusion: Bupleurum n-butanol extract has a protective effect on acute liver injury in mice, and its chemical composition is mainly saikosaponin.
