目的 观察透明质酸酶 (PH 2 0 )基因对人乳腺癌细胞在鸡胚胎中生长和转移的影响。方法 PH 2 0cDNA转染到人乳腺癌细胞株MDA2 31中 (MDA2 31 PH2 0 ) ,细胞种植到鸡胚绒毛尿囊膜 (CAM )上 ,观察肿瘤大小 ;对肿瘤组织血管内皮细胞染色 ,研究肿瘤新生血管生长变化 ;PCR扩增远端CAM中AluDNA序列 ,观察PH | 2 0对肿瘤细胞转移的影响。结果 MDA2 31 PH2 0在CAM上增殖的程度明显高于对照组 ,血管内皮细胞数量也增多 ,同时远端CAM出现AluDNA序列 ,而对照组无。结论 PH 2 0明显促进癌组织内新生血管生长 ,并促进乳癌细胞增殖和远端转移。
Objective To investigate the effect of hyaluronidase (PH 2 0) gene on the growth and metastasis of human breast cancer cells in chicken embryos. Methods The PH 2 0 cDNA was transfected into human breast cancer cell line MDA2 31 (MDA2 31 PH20). The cells were seeded on the chorioallantoic membrane (CAM) of the chick embryo. The tumor size was observed. The tumor endothelial cells were stained and the tumor The growth of neovascularization was observed. The AluDNA sequence in distal CAM was amplified by PCR, and the effect of PH | 20 on the metastasis of tumor cells was observed. Results The proliferation of MDA2 31 PH2 0 in CAM was significantly higher than that in control group, and the number of vascular endothelial cells also increased. At the same time, AluDNA sequence appeared in distal CAM, but not in control group. Conclusion PH 2 0 significantly promote neovascularization in cancerous tissues and promote the proliferation and distant metastasis of breast cancer cells.
