目的:探讨甘肃产华中五味子对大鼠成骨细胞增殖分化的作用。方法:采用血清药理学方法,对体外培养的大鼠成骨细胞作MTT法细胞增殖测定、PNPP法ALP活性测定、以及茜素红染色矿化结节计数。结果:培养48 h,10%、15%和培养72h,5%、10%华中五味子95%乙醇提取物血清组的细胞增殖速度比对照组快(P<0.01);培养48 h,10%、15%和培养72 h,10%华中五味子乙醇提取物血清组的ALP活性较对照组增加(P<0.05和P<0.01);培养2周,矿化结节计数显示10%华中五味子乙醇提取物血清组多于对照组(P<0.01),华中五味子水提取物各血清组对成骨细胞增殖分化没有显著作用。结论:甘肃产华中五味子95%乙醇提取物对体外大鼠成骨细胞的增殖和分化具有促进作用。
Objective: To investigate the effect of Schisandra chinensis from Gansu Province on the proliferation and differentiation of rat osteoblasts. METHODS: Serum pharmacological methods were used for MTT cell proliferation assay, PNPP ALP activity assay, and alizarin red stained mineralized nodule counts for osteoblasts cultured in vitro. RESULTS: After 48 hours of culture, 10%, 15%, and 72 hours of culture, the proliferation rate of 5% and 10% Huazhong Wuweizi 95% ethanol extract sera group was faster than that of the control group (P<0.01); 48 h, 10%, At 15% and 72 hours of culture, the ALP activity of the 10% Huazhong Schisandra ethanol extract sera group was higher than that of the control group (P<0.05 and P<0.01); for 2 weeks, the mineralized nodule count showed 10% Huazhong Schisandra ethanol extract. The serogroup was more than the control group (P<0.01). The schizophrenia water extract of each group had no significant effect on the proliferation and differentiation of osteoblasts. Conclusion: The 95% ethanol extract of Schisandra chinensis from Gansu Province can promote the proliferation and differentiation of rat osteoblasts in vitro.
