目的:探讨通络保肾复方对肾小球足细胞增殖及肾小球足细胞表达nephrin的影响。方法:将5%含药血清培养基加入体外培养的肾小球足细胞,分别于脂多糖(LPS)刺激肾小球足细胞12 h、24 h、36 h、48 h后,MTT方法检测各组肾小球足细胞的增殖情况,以及Western-blot方法检测LPS刺激36 h时肾小球足细胞nephrin的表达。结果:LPS刺激12 h后肾小球足细胞开始增殖,36 h达到高峰,48 h后开始下降。通络保肾复方能够抑制肾小球足细胞的增殖,其中以通络保肾复方大剂量组效果显著,与LPS组比较差异有统计学意义(P<0.05)。同时通络保肾复方可以上调36 h时由LPS刺激导致的肾小球足细胞nephrin的表达。结论:本研究结果提示,通络保肾复方可以抑制LPS诱导的肾小球足细胞的增殖,上调肾小球足细胞nephrin的表达,减少足细胞的损伤,降低蛋白尿的发生,从而延缓糖尿病肾病的进一步发展。
Objective: To investigate the effect of Tonglu Baoshen Recipe on glomerular podocyte proliferation and nephrin expression in glomerular podocytes. Methods: 5% serum-containing medium was added into glomerular podocytes cultured in vitro, respectively. Glomerular podocytes were stimulated with lipopolysaccharide (LPS) for 12 h, 24 h, 36 h, 48 h, Group glomerular podocyte proliferation, and Western-blot method to detect glomerular podocyte nephrin expression 36 h after LPS stimulation. RESULTS: Glomerular podocytes began to proliferate after 12 h of LPS stimulation, reached a peak at 36 h, and then decreased at 48 h. Tongluo Baogan Decoction can inhibit the proliferation of glomerulus podocytes, among which Tongluoluxin Decoction high dose group has significant effect, compared with LPS group, the difference is statistically significant (P <0.05). At the same time, Tongluo Baoshen Compound can up-regulate glomerular podocyte nephrin expression induced by LPS at 36 h. Conclusion: The results of this study suggest that Tongluo Baogan Decoction can inhibit the proliferation of glomerular podocytes induced by LPS, up-regulate the expression of nephrin in glomerular podocytes, reduce the damage of podocytes, reduce the occurrence of proteinuria and delay the progression of diabetes mellitus Nephropathy further development.
