以蛹虫草为试材,研究蛹虫草生长过程中子实体鲜重、培养基pH值、胞外酸性蛋白酶、中性蛋白酶和碱性蛋白酶的活性变化。结果表明:蛹虫草酸性蛋白酶的活性大小受氮源的影响,由112U/g提高至213U/g,但形成规律基本一致。胞外中性和碱性蛋白酶的活性高低及形成规律均受到氮源的影响。中性蛋白酶活性在氮源培养基中第30天达到最高峰,为10.2U/g,而在空白培养基上却未检测得显著的峰值。碱性蛋白酶活性受氮源的影响在第10天开始上升,至第50天达到最大值(10U/g)。在空白培养基上,碱性蛋白酶活性在第50天开始上升,培养结束时达最大值(6.5U/g)。这3种蛋白酶中,酸性蛋白酶活性与子实体的生长趋势相关性最大。该研究将为蛹虫草子实体生产的营养生理研究提供参考依据。
Using Cordyceps militaris as test material, the changes of fresh weight of fruiting bodies, pH value, extracellular acidic protease, neutral protease and alkaline protease in the growth of Cordyceps militaris were studied. The results showed that the activity of Cordyceps militaris acid protease increased from 112 U / g to 213 U / g under the influence of nitrogen source, but the formation law was basically the same. The activity of extracellular neutral and alkaline protease and the formation of the law are affected by the nitrogen source. Neutral protease activity peaked at day 30 in the nitrogen-source medium at 10.2 U / g, whereas no significant peak was detected on the blank medium. Alkaline protease activity began to rise on day 10 due to the effect of nitrogen sources and reached a maximum at day 50 (10 U / g). On blank medium, alkaline protease activity began to rise on the 50th day and reached the maximum at the end of the culture (6.5 U / g). Of the three proteases, the activity of acid protease was most closely related to the growth of fruiting bodies. This research will provide reference for the nutrition and physiology of Cordyceps militaris production.
