目的:研究皮质醇对激素依赖型乳腺癌细胞MCF-7增殖、迁移的影响及其可能的作用机制。方法:不同浓度皮质醇处理激素依赖型人乳腺癌细胞株MCF-7。采用MTS法检测不同浓度皮质醇对细胞增殖的影响;细胞划痕试验检测不同浓度皮质醇对细胞迁移的影响;蛋白质印迹法检测细胞增殖相关蛋白磷酸化m TOR(p-m TOR)以及迁移相关蛋白MMP-9、MMP-2蛋白表达变化情况。结果:MCF-7细胞在皮质醇剂量为10 nmol/L时增殖显著(P<0.05)、迁移增加(P<0.01)。并且在皮质醇剂量10 nmol/L时,MCF-7细胞表达细胞增殖相关蛋白p-m TOR(P<0.05),及细胞迁移相关蛋白MMP-9(P<0.05)、MMP-2(P<0.01)明显升高,差异具有统计学意义。结论:小剂量(10 nmol/L)皮质醇可以导致激素依赖型乳腺癌细胞增殖、迁移增加,其机制可能是通过激活经典m TOR通路诱导细胞增殖及上调MMP-9、MMP-9蛋白表达实现。
Objective: To investigate the effects of cortisol on the proliferation and migration of hormone-dependent breast cancer cell line MCF-7 and its possible mechanism. Methods: Hormone-dependent human breast cancer cell line MCF-7 was treated with different concentrations of cortisol. The effects of different concentrations of cortisol on cell proliferation were detected by MTS assay; the effects of different concentrations of cortisol on cell migration were detected by cell scratch assay; the expressions of mTOR (pm TOR) and migration-associated protein MMP -9, MMP-2 protein expression changes. Results: The proliferation of MCF-7 cells was significantly increased (P <0.05) and the migration was increased when the cortisol dose was 10 nmol / L (P <0.01). (P <0.05), MMP-9 (P <0.05) and MMP-2 (P <0.01) in MCF-7 cells at cortisol dose of 10 nmol / Significantly increased, the difference was statistically significant. CONCLUSION: Small dose (10 nmol / L) of cortisol can induce the proliferation and migration of hormone-dependent breast cancer cells. The mechanism may be through activating the classical mTOR pathway to induce cell proliferation and up-regulating the expression of MMP-9 and MMP-9 .
