目的探讨顺铂联合葡萄籽原花青素对宫颈癌Hela细胞的生长抑制作用。方法 2015年10月—2016年1月体外培养宫颈癌Hela细胞,分为对照组、顺铂组、原花青素组及顺铂联合原花青素组,48、72 h后,倒置显微镜下观察细胞形态学变化,CCK8法检测细胞增殖抑制率。三组及以上计量资料比较采用单因素方差分析,两两比较采用LSD法;计数资料比较采用χ2检验,P<0.05为差异有统计学意义。结果顺铂及原花青素对Hela细胞的增殖抑制呈现剂量和时间相关性,随着药物浓度的升高和作用时间的延长,Hela细胞的增殖抑制率升高。联合用药组9、18、37μmol/ml的顺铂联合340μmol/ml的原花青素分别作用48 h后Hela细胞的增殖抑制率分别为75.7%、76.1%、76.2%,72 h后Hela细胞的增殖抑制率分别为77.0%、78.0%、78.9%。两者联合用药组细胞增殖抑制率高于单独用药组,差异均有统计学意义(均P<0.05)。结论不同浓度顺铂、原花青素对Hela细胞增殖均有抑制作用,原花青素对Hela抑制作用显著,顺铂联合原花青素对宫颈癌细胞Hela有协同抑制作用。
Objective To investigate the inhibitory effect of cisplatin combined with grape seed proanthocyanidin on the growth of cervical cancer Hela cells. Methods Hela cells were cultured in vitro from October 2015 to January 2016 and divided into control group, cisplatin group, proanthocyanidin group and cisplatin combined with proanthocyanidins group. 48 and 72 hours later, morphological changes were observed under inverted microscope, CCK8 assay for inhibition of cell proliferation. Three groups and above measurement data were compared using one-way ANOVA, compared with each other by LSD method; count data were compared using χ2 test, P <0.05 for the difference was statistically significant. Results Cisplatin and proanthocyanidin inhibited the proliferation of Hela cells in dose-and time-dependent manner. With the increase of drug concentration and prolongation of action time, the proliferation inhibition rate of Hela cells increased. The inhibitory rates of proliferation of Hela cells treated with 9, 18, 37μmol / ml cisplatin combined with 340μmol / ml proanthocyanidin for 48 h were 75.7%, 76.1%, 76.2% and 72 h, respectively. Respectively 77.0%, 78.0% and 78.9%. The inhibitory rate of cell proliferation between the combination group was higher than that of the drug alone group (all P <0.05). Conclusion Different concentrations of cisplatin and proanthocyanidin have inhibitory effects on the proliferation of Hela cells. Proanthocyanidins inhibit Hela significantly. Cisplatin combined with proanthocyanidins have synergistic inhibitory effect on Hela cells.
