目的建立大鼠血浆中杨梅素浓度的分析方法。方法采用UPLC-MS法对大鼠血浆中的杨梅素进行含量测定。结果杨梅素浓度在10-4 000 ng.mL-1内线性关系良好;定量下限为10 ng.mL-1;低(20 ng.mL-1)、中(200 ng.mL-1)、高(4000 ng.mL-1)三个浓度的相对回收率分别为(102.81±6.54)%、(109.82±3.94)%和(105.79±2.06)%,提取回收率分别为(41.70±6.70)%、(39.52±2.67)%和(47.87±1.49)%;基质效应分别为(97.31±2.22)%、(96.31±3.09)%和(110.07±4.98)%;三个浓度的日内、日间精密度RSD均在5%以内。结论本方法准确可靠、简便快速,适用于大鼠血浆中杨梅素浓度的测定。
Objective To establish a method for the analysis of myricetin in rat plasma. Methods The content of myricetin in rat plasma was determined by UPLC-MS. Results The myritin concentration was linear in the range of 10-4 000 ng.mL-1 with a lower limit of quantitation of 10 ng.mL-1, low (20 ng.mL-1), medium (200 ng.mL-1) The relative recoveries were (102.81 ± 6.54)%, (109.82 ± 3.94)% and (105.79 ± 2.06)%, respectively, and the recovery rates were (41.70 ± 6.70)% and (39.52 ± 2.67)% and (47.87 ± 1.49)% respectively. The matrix effects were (97.31 ± 2.22)%, (96.31 ± 3.09)% and (110.07 ± 4.98)%, respectively. The intra- and inter- Are less than 5%. Conclusion The method is accurate, reliable, simple and rapid and suitable for the determination of myricetin in rat plasma.
