目的:探讨绿色荧光蛋白(GFP)在环磷酰胺(CY)处理小鼠胸腺中的表达。方法:将质粒pEGFP-c3和梭华SofastTM基因转染试剂按一定比例配制成转染液,经腹腔注入3组小鼠(每组15只)进行转染。转染24h后分别注射0.5、1、2mg/只的CY,并设对照组(15只)。18h后称小鼠体质量,然后处死动物,无菌取胸腺并称重,用PBS洗出细胞,制成单细胞悬液,于荧光显微镜下观察GFP的表达情况。结果:GFP可在小鼠胸腺中表达,且GFP+细胞数与CY的剂量相关。高剂量的CY处理可导致胸腺明显萎缩、胸腺细胞总数及GFP+细胞数的减少。结论:转染质粒pEGFP-c3并经CY处理的小鼠及正常对照组小鼠的胸腺中均可表达GFP,但CY处理的小鼠胸腺中GFP的表达情况明显低于对照组小鼠。
Objective: To investigate the expression of green fluorescent protein (GFP) in the thymus of cyclophosphamide (CY) -treated mice. Methods: Plasmid pEGFP-c3 and SofastTM transfection reagent were prepared into transfection solution according to a certain proportion. Three groups of mice (15 in each group) were injected intraperitoneally for transfection. Transfection 24h after injection of 0.5, 1, 2mg / only CY, and control group (15). After 18h, the mice were weighed and the animals were sacrificed. The thymus glands were aseptically taken and weighed. The cells were washed with PBS to make a single cell suspension. The expression of GFP was observed under a fluorescence microscope. Results: GFP was expressed in the mouse thymus, and the number of GFP + cells was related to the dose of CY. High doses of CY treatment resulted in significant atrophy of the thymus, total thymus cells, and a decrease in GFP + cells. CONCLUSION: GFP can be expressed in the thymus of mice treated with pEGFP-c3 and CY treated and normal control mice, but the expression of GFP in CY treated mice is significantly lower than that of the control mice.
