目的探讨人细粒棘球蚴感染中Tim-3/Galectin-9与Thl/Th2的平衡关系及其与该疾病发生、发展的关系。方法用实时荧光定量PCR法分别检测30例慢性细粒棘球蚴感染者和30例健康体检人群外周血中Tim-3、Galectin-9、T-bet和GATA-3mRNA的表达,同时用酶联免疫吸附(ELISA)法检测慢性细粒棘球蚴感染者和健康对照者血清中可溶性Tim-3和Galectin-9及IFN-γ、IL-12和IL-4的水平。结果 (1)实时荧光定量PCR检测结果显示:与健康对照组相比,感染组Tim-3和Galectin-9mRNA水平显著升高,T-bet mRNA水平显著降低,GATA-3mRNA水平显著升高。(2)ELISA检测结果显示:与健康对照组相比,感染组血清中可溶性Tim-3和Galectin-9水平显著升高,IFN-γ和IL-12水平显著下降,IL-4水平显著升高。Tim-3与Galectin-9呈正相关(P=0.000,r=0.776);Tim-3与T-bet呈负相关(P=0.000,r=-0.645);Tim-3与GATA-3呈正相关(P=0.000,r=-0.752)。结论 Tim-3/Galectin-9在人细粒棘球蚴持续性感染中发挥一定的作用,Tim-3/Galectin-9可能通过影响Th1/Th2相关的转录因子T-bet/GATA-3和细胞因子IL-12、IFN-γ和IL-4的水平变化,促进Th1/Th2细胞失衡现象的发生,从而促进感染的持续性发展。
Objective To investigate the equilibrium relationship between Tim-3 / Galectin-9 and Thl / Th2 in human Echinococcus granulosus infection and its relationship with the occurrence and development of this disease. Methods The expression of Tim-3, Galectin-9, T-bet and GATA-3 mRNA in peripheral blood of 30 patients with chronic echinococcosis and 30 healthy controls were detected by real-time fluorescence quantitative PCR. Serum levels of soluble Tim-3 and Galectin-9, IFN-γ, IL-12 and IL-4 were measured by ELISA in patients with chronic echinococcosis and healthy controls. Results (1) The results of real-time PCR showed that compared with the healthy control group, the levels of Tim-3 and Galectin-9 mRNA in the infected group were significantly increased, the levels of T-bet mRNA and GATA-3 mRNA were significantly increased. (2) The results of ELISA showed that the levels of soluble Tim-3 and Galectin-9 in serum were significantly increased, the levels of IFN-γ and IL-12 were significantly decreased and the levels of IL-4 were significantly increased . Tim-3 was positively correlated with Galectin-9 (P = 0.000, r = 0.776); Tim-3 was negatively correlated with T-bet P = 0.000, r = -0.752). Conclusion Tim-3 / Galectin-9 plays a role in the persistent infection of human Echinococcus granulosus. Tim-3 / Galectin-9 may affect Th1 / Th2 related transcription factors T-bet / GATA- The changes of IL-12, IFN-γ and IL-4 levels promote the imbalance of Th1 / Th2 cells and promote the continuous development of infection.
