目的探讨六经头痛片治疗偏头痛的作用及可能的机制。方法通过sc硝酸甘油复制大鼠偏头痛模型,观察大鼠搔头次数和搔头反应潜伏期;采用酶联免疫实验(ELISA)检测脑组织中5-羟色胺(5-HT)、去甲肾上腺素(NE)、多巴胺(DA)水平及血清中β-内啡肽(β-EP)、降钙素基因相关肽(CGRP)、内皮素(ET)的量,采用生化法测定血清中一氧化氮(NO)量及一氧化氮合酶(NOS)活性。结果模型组大鼠在sc硝酸甘油后1~3 min即出现搔头,且次数频繁。与模型组比较,六经头痛片1.4、0.7 g/kg组可显著减少偏头痛模型大鼠的搔头次数(P<0.001),并呈剂量相关性,能够明显延长大鼠搔头反应的潜伏期(P<0.01);六经头痛片1.4、0.7 g/kg组可显著升高模型大鼠血清β-EP、ET及脑内DA的量,显著降低血清CGRP的量和NOS活性,六经头痛片1.4 g/kg组可明显降低血清NO的量,对脑内5-HT、NE的量基本无影响。结论六经头痛片有显著的治疗偏头痛作用,可以升高血清β-EP、ET、DA的量,降低CGRP、NO的量及NOS活性,其作用机制与收缩血管及中枢镇痛有关。
Objective To explore the effect and possible mechanism of Liujingtountong Tablet on migraine. Methods Rat migraine model was replicated by nitroglycerin (nitroglycerin), and the frequency of scratching and the latency of scratching were observed. The levels of 5-hydroxytryptamine (5-HT), norepinephrine (NE), dopamine (DA), β-EP, CGRP and ET in serum were measured by enzyme-linked immunosorbent assay NO) and nitric oxide synthase (NOS) activity. Results In the model group, scrotum appeared frequently 1 ~ 3 minutes after sc nitroglycerin. Compared with the model group, Liuutouotong tablets 1.4,0.7 g / kg group can significantly reduce the frequency of scratching headache in migraine model rats (P <0.001), and in a dose-dependent manner, can significantly prolong the latency of the rat’s scratching reaction (P <0.01). Liuutuo Tongliao Tablet (1.4,0.7 g / kg) could significantly increase the levels of serum β-EP, ET and brain DA in model rats, and significantly reduce the serum levels of CGRP and NOS. 1.4 g / kg tablet can significantly reduce the amount of serum NO, the amount of brain 5-HT, NE no significant effect. Conclusions Liujingtou Pian has significant effect on migraine, which can increase the levels of serum β-EP, ET and DA, and decrease the levels of CGRP, NO and NOS. The mechanism of action is related to the vasoconstriction and central analgesia.
