目的 :探讨凋亡蛋白抑制因子 (IAP)SurvivinmRNA及蛋白在人脑胶质瘤细胞系BT32 5和SHG 4 4中的表达水平及意义 .方法 :采用反转录聚合酶链反应 (RT PCR)和免疫组化方法分别检测SurvivinmRNA及蛋白在BT32 5和SHG 4 4细胞系中的表达水平 .结果 :BT32 5细胞高表达SurvivinmRNA及蛋白 ,并表达少量的Survivin剪接变异体Survivin 2B ,Survivin蛋白定位于瘤细胞的胞质 .SurvivinmRNA及蛋白在SHG 4 4细胞中均无表达 .结论 :Survivin对于维持BT32 5细胞株的生物学特性具有重要作用 ;BT32 5和SHG 4 4细胞株可分别作为Survivin基因抑制和基因转移研究的良好细胞模型 .
Objective: To investigate the expression and significance of Survivin mRNA and protein of apoptosis inhibitor protein (IAP) in human glioma cell lines BT32 5 and SHG 4. Methods: Reverse transcription polymerase chain reaction (RT PCR) and Immunohistochemistry was used to detect the expression of Survivin mRNA and protein in BT32 5 and SHG 4 4 cell lines respectively.Results: Survivin mRNA and protein were highly expressed in BT32 5 cells and a small amount of Survivin splicing variants Survivin 2B and Survivin protein were localized in the tumor Cytosolic cytoplasm.Survivin mRNA and protein were not expressed in SHG 4 4 cells.Conclusion: Survivin plays an important role in maintaining the biological characteristics of BT32 5 cell line; BT32 5 and SHG 4 4 cell lines can be used as Survivin gene inhibition and Good cell model for gene transfer studies.
