目的 :观察大豆皂甙对高浓度胰岛素下兔血管平滑肌细胞脂质过氧化反应的影响。方法 :采用兔主动脉平滑肌细胞培养 ,细胞随机分为正常对照组、胰岛素组及不同浓度大豆皂甙组 ,加药后继续培养 72 h,计数后测定细胞内过氧化脂质 (LPO)及超氧化物歧化酶 (SOD)含量。结果 :单纯高浓度胰岛素升高细胞内 LPO水平 ,降低细胞内 SOD活力 ,大豆皂甙与胰岛素共同孵育后 ,升高SOD活力 ;大豆皂甙低于 2 0 mg/L时抑制 LPO的产生 ,高于或等于 50 mg/L时促进 LPO的合成。结论 :高浓度胰岛素通过升高平滑肌细胞内 LPO促进动脉粥样硬化的形成 ,一定浓度的大豆皂甙抑制胰岛素所致脂质过氧化反应 ,对动脉粥样硬化具有防治作用
Objective: To observe the effect of soybean saponin on lipid peroxidation of rabbit vascular smooth muscle cells under high concentration of insulin. Methods: Rabbit aortic smooth muscle cells were cultured. The cells were randomly divided into normal control group, insulin group and different concentrations of soybean saponin group. The cells were cultured for 72 h. After counting, the levels of intracellular lipid peroxide (LPO) and superoxide Dismutase (SOD) content. Results: High concentrations of insulin alone increased intracellular LPO levels and decreased intracellular SOD activity. Soybean saponin incubated with insulin increased SOD activity. Soybean saponins inhibited LPO production below 20 mg / L, Equal to 50 mg / L to promote the synthesis of LPO. CONCLUSION: High concentration of insulin enhances the formation of atherosclerosis by increasing LPO in smooth muscle cells. A certain concentration of soyasaponin inhibits insulin-induced lipid peroxidation and has a preventive and therapeutic effect on atherosclerosis
