卵母细胞在冷冻保护剂加载过程中受到的渗透损伤和毒性损伤会影响细胞的存活率,针对这一情况,通过软件模拟来了解卵母细胞在不同冷冻保护剂加载情况下的渗透行为,以寻求合适的加载方案使得卵母细胞所受的损伤最小。利用MLAB编程分别模拟出卵母细胞在四步法、一般线性法、凸型线性法以及凹型线性法加载冷冻保护剂的方案下的渗透行为;综合积累性渗透损伤AOD、积累性毒性损伤J、归一化体积V/V0三项参数来评估卵母细胞受到的损伤。结果表明,当胞外冷冻保护剂为乙二醇时,采用凹型线性加载法加载冷冻保护剂对卵母细胞造成的损伤最小。
