探讨联用褐藻胶降解物(ADP)与头孢类抗生素对临床常见耐药菌的抑制活性。在不同条件下酸降解褐藻胶得到ADP-1、ADP-2、ADP-3和ADP-4,分别与6种头孢类抗生素联用。以耐药的铜绿假单胞菌、金黄色葡萄球菌、大肠埃希菌,以及泛耐药的鲍曼不动杆菌为受试对象,采用96孔板倍比稀释法进行抑菌活性筛选,通过计算抑菌率评价ADP与头孢类抗生素联合抑制耐药菌的效果。结果表明4种ADP与头孢类抗生素联用后,抑菌率大多提高到80%以上,尤其是头孢拉定与ADP-3联用后,对耐药铜绿假单胞菌的抑制率提高了116倍。结果提示ADP可进一步开发成为抗菌增效药物。
To investigate the inhibitory activity of combined use of alginate degradation products (ADP) and cephalosporins on clinical resistant strains. ADP-1, ADP-2, ADP-3 and ADP-4 were obtained by acid-degrading alginate under different conditions and were respectively combined with 6 kinds of cephalosporins. To drug-resistant Pseudomonas aeruginosa, Staphylococcus aureus, Escherichia coli, and Pan-resistant Acinetobacter baumannii as the test object, using 96-well plate dilution ratio antibacterial activity screening by Antibacterial rate was calculated to evaluate the effect of ADP combined with cephem antibiotics in inhibiting drug-resistant bacteria. The results showed that the combination of four kinds of ADP and cephalosporin antibiotics, the antibacterial rate increased mostly to more than 80%, especially with cefradine and ADP-3 combination, the resistance of resistant Pseudomonas aeruginosa increased by 116 times. The results suggest that ADP can be further developed into an antimicrobial synergistic drug.
