目的研究α-干扰素和异博定对体外培养的乳腺癌细胞多药耐药的逆转作用。方法MTT法检测各实验组细胞存活率,计算IC50、耐药倍数和逆转倍数;流式细胞术定量检测细胞表面P170的表达。结果α-干扰素与异博定联合应用使乳腺癌细胞耐阿霉素(ADM)的IC50降低为0.32μmol/L,优于单用异博定(1.23μmol/L)、α-干扰素(2.29μmol/L),逆转倍数由7.25和13.49提高到51.88;且P-170表达均有大幅度下降。结论单独用α-干扰素、异博定均可达到部分逆转乳腺癌耐药细胞(MCF-7/ADR)对ADM的耐药的作用,二者联合使用效果更强。
Objective To study the reversal effects of α-interferon and ibuprofen on multidrug resistance in cultured breast cancer cells in vitro. Methods The cell viability was measured by MTT assay, and the IC50, the multiple of drug resistance and the multiple of drug resistance were calculated. The expression of P170 protein was detected by flow cytometry. Results The combination of α-interferon and ibuprofen reduced the IC50 of adriamycin (ADM) to 0.32 μmol / L in breast cancer cells, which was better than that of ibotidine (1.23 μmol / L), α-interferon 2.29μmol / L). The fold of reversal increased from 7.25 and 13.49 to 51.88, respectively. The expression of P-170 decreased significantly. Conclusions Both interferon and isoborne alone can partially reverse the resistance of ADM-resistant breast cancer cells (MCF-7 / ADR) to ADM.
