目的:研究天麻酚性成分对羟基苯甲醇(4-hydroxybenzyl alcohol)对血脑屏障的保护作用。方法:雄性大鼠随机分为假手术组、模型组、对羟基苯甲醇20mg/kg组、对羟基苯甲醇10mg/kg组。采用线栓法复制大鼠脑缺血再灌注(MCAO/R)模型,通过神经学评分观察大鼠神经功能缺失症状;采用伊文思蓝渗透法观察血脑屏障通透性;运用Western Blot技术检测血脑屏障相关蛋白AQP-4、Claudin-5及Occludin的表达。结果:大鼠脑缺血2h再灌注24h后,对羟基苯甲醇20mg/kg、10mg/kg均对模型大鼠的神经功能缺失症状具有改善作用,能够降低缺血侧脑组织中伊文思蓝的含量,降低AQP-4蛋白的表达,对紧密连接蛋白(Claudin-5、Occludin)的表达无明显影响。结论:天麻酚性成分对羟基苯甲醇对大鼠脑缺血再灌注损伤后血脑屏障具有保护作用,作用机制可能与其降低AQP-4蛋白的表达有关。
Objective: To study the protective effects of 4-hydroxybenzyl alcohol on the blood-brain barrier of phenolic compounds in Gastrodia elata Blume. Methods: Male rats were randomly divided into sham-operation group, model group, p-hydroxybenzyl alcohol 20 mg / kg group and p-hydroxybenzyl alcohol 10 mg / kg group. The rat model of cerebral ischemia-reperfusion (MCAO / R) was established by the method of thread occlusion. The neurological deficits were observed by neurological score. The permeability of blood-brain barrier was observed by Evans blue permeation method. The expression of blood brain barrier related protein AQP-4, Claudin-5 and Occludin. Results: After 2 h reperfusion in cerebral ischemia rats, p-hydroxybenzyl alcohol 20 mg / kg and 10 mg / kg all improved the neurological deficits in the model rats and decreased the expression of Evans blue Content, decreased the expression of AQP-4 protein, and had no significant effect on the expression of Claudin-5 (Occludin). CONCLUSION: The phenolic compounds of Gastrodia elata Blume have a protective effect on the blood-brain barrier after cerebral ischemia-reperfusion injury in rats. The mechanism may be related to the decrease of AQP-4 protein expression.
