通过内路法将分离后的有色兔RPE细胞移植到无色兔眼的视网膜下腔,共完成10只兔(20只眼)的实验手术,分术后2天(2只眼)、10天(6只眼)、20天(6只眼)、40天(4只眼)及90天(2只眼)5个不同观察组,结果表明移植的RPE细胞2天后未完全贴伏于Bruch膜上,术后10天则紧密贴伏其上并形成单细胞层。术后40天移植的RPE细胞与视网膜神经层形成相嵌结构。术后90天移植细胞内有吞噬消化的外节盘膜,提示移植细胞正在行使其新陈代谢的功能。RPE移植成功,为进一步研究移植RPE细胞的生长、功能及免疫反应,并为临床应用奠定基础。
The isolated rabbit RPE cells were transplanted into the subretinal space of colorless rabbit eyes by intra-procedural method. A total of 10 rabbits (20 eyes) were performed experimental surgery, 2 days (2 eyes), 10 days (6 eyes), 20 days (6 eyes), 40 days (4 eyes) and 90 days (2 eyes). The results showed that the transplanted RPE cells did not completely adhere to Bruch’s membrane after 2 days On the 10th postoperative day, they were closely attached to and form a single cell layer. RPE cells transplanted 40 days after surgery with the retinal nerve layer to form phase-embedded structure. The phagocytosed digestive outer membrane was transplanted in the transplanted cells 90 days after operation, suggesting that the transplanted cells are exercising their metabolic function. RPE transplantation was successful, in order to further study the growth, function and immune response of transplanted RPE cells and lay the foundation for clinical application.
