Objective To observe the effects of triptolide on the peripheral blood γδT cell ratio and apoptosis in type II collagen-induced arthritis (CIA) rats, and to explore the possible mechanism of Tripterygium wilfordii treatment for rheumatoid arthritis (RA). Methods The rat model of arthritis induced by collagen was used as animal model. Twenty rats with successful modeling were randomly divided into model group and triptolide group. Triptolide was intramuscularly administered at 4 μg/100 g body weight. Animals were sacrificed every 3 days for 31 days. The ratio of γδ T cells was measured by direct immunofluorescence, and the apoptosis rate of γδT cells was measured by flow cytometry. Results Compared with the normal control group, the ratio of γδT cells in the peripheral blood of the CIA model group decreased significantly and the apoptotic rate increased (P<0.01). After treatment with triptolide, the ratio of γδT cells recovered and approached the normal control group. The difference was not statistically significant, P>0.05. Conclusion The reduction of γδT cell apoptosis and the regulation of the ratio of γδ T cells in peripheral blood of CIA rats may be one of the mechanisms of triptolide in the treatment of RA.
