目的观察洛铂对胃癌细胞SGC-7901细胞增殖的影响,并探讨其作用机制。方法将对数生长期的SGC-7901细胞分阴性对照组(不加药物)和实验组(加入不同浓度洛铂),采用倒置显微镜观察不同浓度下细胞生长状态;MTT法观察对细胞的增殖抑制;Western印迹方法分析不同浓度洛铂对SGC-7901细胞内Bcl-2蛋白表达水平的影响。结果倒置显微镜下观察,洛铂能抑制细胞增殖,出现明显的凋亡学形态特征。MTT法显示洛铂能抑制细胞增殖,并呈浓度及时间依赖关系。Western印迹结果显示洛铂能使SGC-7901细胞Bcl-2蛋白表达水平下降。结论洛铂能明显抑制胃癌SGC-7901细胞增殖,诱导凋亡,可能与调节SGC-7901细胞Bcl-2蛋白表达水平相关。
Objective To observe the effect of lobaplatin on the proliferation of gastric cancer cell line SGC-7901 and to explore its mechanism. Methods SGC-7901 cells in logarithmic growth phase were divided into negative control group (untreated group) and experimental group (treated with different concentrations of lobaplatin). The growth of cells in different concentrations was observed by inverted microscope. The proliferation inhibition The effect of different concentrations of lobaplatin on the expression of Bcl-2 protein in SGC-7901 cells was analyzed by Western blotting. Results Under the inverted microscope, lobaplatin inhibited cell proliferation and showed obvious apoptotic morphological features. MTT assay showed that lorazepam inhibited cell proliferation in a concentration- and time-dependent manner. Western blotting showed that Losalab decreased the expression of Bcl-2 protein in SGC-7901 cells. Conclusion Losopin can significantly inhibit the proliferation and induce apoptosis of gastric cancer SGC-7901 cells, which may be related to the regulation of Bcl-2 protein expression in SGC-7901 cells.
